The Department also houses a 1200 sq. ft. suite to support protein expression and purification from different organisms ranging from bacteria to mammalian cells. The facility has four separate rooms and necessary instrumentation to perform protein expression in bacteria, yeast, insect cells, and mammalian cells with a cold room. Two large refrigerated environmental shakers are designated for bacterial and yeast culture work separately in each room. A 7.5L fermentor is also available. For insect and mammalian cell work, each room is equipped with a laminar air-flow workstation, microscope, water bath, low-speed centrifuge, cell counter, and CO2 incubators. For large culture work, a 5L bioreactor is also available. A low speed centrifuge with 6L capacity and super speed centrifuge are located in a shared room. Protein purification equipment and instruments for biophysical characterization of proteins are available as well. They include AKTAXpress twin and AKTA explorer units. A UV-VIS spectrometer and single cuvette port and scanning fluorescence plate readers are housed in the facility. Analytical instruments comprise a circular dichroism spectrometer with six position Peltier thermoelectric type temperature control, differential scanning calorimeter, isothermal titration calorimeter, spectrofluorometer, Biacore 3000, and a multi-angle light scattering system. In addition we have a walk-up access to the mass spec instrumentation in the proteomics core laboratory in BST3. For high resolution top-down and bottom up proteomics work we have a FT MS with 12 Tesla magnet available for determination of molecular weights and sequences of polypeptide mixtures with highest accuracy.
